Molecular biology laboratory protocols: plant genotyping

This training manual describes the following protocols for plant genotyping, all used successfully for potatoes, sweetpotatoes, and Andean roots and tubers at CIP's molecular biology laboratory:

  • Medium- and small-scale plant DNA extraction;
  • Mini-scale plasmid extraction;
  • PCR amplification of plasmid inserts;
  • Non-radioactive RFLP analysis;
  • Random amplified polymorphic DNA (RAPD);
  • Single sequence repeats (SSR) or microsatellites;
  • Amplified fragment length polymorphism (AFLP);
  • Non-radioactive amplified fragment length polymorphism;
  • DNA gel electrophoresis; and
  • Purification of TAQ polymerase (short protocol).
